Understanding Slavery Essay A poignantly moving tale of a womanââ¬â¢s courage and determination in the face of seemingly insurmountable odds, Incidents in the Life of a Slave Girl undoubtedly serves as an inspiration for those who endeavor to rise beyond their initial station in life on the way to achieving oneââ¬â¢s dreams. Though the author claims it to be a historical account, it could easily pass off for a work of fiction in the tradition of the historical novel ââ¬â a romanticizing American life and history with its tale of noble suffering, heroic daring and unwavering zeal. Perhaps more significantly, its merit lies in offering the reader the painful truth of the slave experience through the eyes of an African-American slave girl. As Jacobs narrates, she was born a slave but she never knew it until six years of happy childhood had passed away (Jacobs, p. 1). Though they were all slaves in the family, she was so fondly shielded from that fact and they appear to have lived normal lives that she never dreamed she was ââ¬Å"a piece of merchandise trusted to them for safe keeping, and liable to be demanded of them at any momentâ⬠(Jacobs, p. 1). That is, until her mother died and her life took a turn for the worse, experiencing first hand what it meant to be a slave in those days. Because of the way she was raised, it would appear that Linda Brent is exceptional for a woman of her social stature. Having received the rudiments of a basic education at the hands of her familyââ¬â¢s original owners, she is clear thinking and possesses a keen intellect. Not surprisingly, she was able to transcend the limitations of race, class and gender and grasp the reality that women, regardless of color, race and stature, share the common experience of victimization at the hands of a male-dominated, patriarchal society. Moreover, by virtue of the loving relationships she had established early on with her family and immediate community, she is quite capable of empathy. Her initial understanding of the nature of slavery, though nonetheless negative, did not prevent her from maintaining a positive outlook towards life, notwithstanding her position in the social hierarchy. This however has been severely challenged throughout the years of her ordeal. Due to the many betrayals she had experienced in her lifetime, she learns to distrust people, and even though this is gradually tempered by the formation of positive relationships, she retains her hesitant, guarded approach to life. The bookââ¬â¢s main strength lies in its rich narratives, the vivid portrayal of the female slave experience, and its use of the female point of view in the narration of her tale. In terms of limitations, the book would have problems in its illustration of the slave experience as representative of the larger Africa-America black community. Linda Brent and her family could be considered among the lucky few fate seemed to have favored among the millions more of their kind, who, though some might have endeavored to attain their freedom were faced with more dire, less relenting circumstances which made freedom remain a dream for them. A cursory glance at historical accounts of slavery would reveal that many have attempted to escape from their masters, with a considerable number dying in the attempt. In Jacobsââ¬â¢ particular tale, the experience of women in bondage is illuminated. Yet more than a narration of physical infliction of pain, torture and misery ââ¬â the often-told tale of American slavery ââ¬â what is striking is how she makes the reader comprehend that the most devastating blow endured and inflicted upon female slaves is that of the continuing existence of a system recognizing, even illicitly sanctioning concubinage and licentiousness among white males, the double standard of the times which make it doubly hard for women, most especially slaves, to live a decent, dignified existence wherein they could realize their true worth as individuals. For the slaves in Americaââ¬â¢s colonial past, every day was deplorable. Suffering in its various forms ââ¬â physical, mental, emotional, psychological and spiritual ââ¬â was common-day fare subject to the wiles and dispositions of their masters. For both men and women, manual labor, e. g. working the fields in the plantations of the South, was a large component of the day. As Jacobs (p. 12) illustrates: ââ¬Å"On a farm, they work until the corn and cotton are laid. They then have two holidays. Some masters give them a good dinner under the trees. This over, they work until Christmas Eve. If no heavy charges are meantime brought against them, they are given four or five holidays, whichever the master or overseer may think proper. â⬠For the women, this was often aggravated by (more often than not) unwarranted sexual advances, if not from their masters, the other male members of the household, or among fellow slaves. New Yearââ¬â¢s Day was a terribly appalling affair, for hiring day at the South took place every first of January. To the slave mother, New Years Day comes laden with peculiar sorrows: ââ¬Å"She sits on her cold cabin floor, watching the children who may all be torn from her the next morning; and often does she wish that she and they might die before the day dawns. She may be an ignorant creature, degraded by the system that has brutalized her from childhood; but she has a mothers instincts, and is capable of feeling a mothers agoniesâ⬠(Jacobs, p. 13). Children born as slaves were sold off at the auction block, for they belonged to their master just as their parents did, for him to do with as he pleases. For is that not the nature of property? Slaves were treated as such, not regarded as human beings, but rather little more than animals to be set to labor upon the fields, to assist in the keeping of the household, to run errands and perform manual labor deemed unworthy of the white masterââ¬â¢s unsoiled hands. Those same white hands were quick with the whip for every transgression committed by an erring slave, and a mouth which reserved the foulest of words to further degrade the slave and instill in his/her consciousness his/her lack of worth, how inferior and far beneath their master they were. The ensuing slave consciousness formed from this inhuman treatment, nourished throughout a life of bondage and suffering, is bitter and bleak, their minds ignorant and uncultured, deprived of the conditions which give human existence its dignity, the individual his self-worth (Fowler and Fowler, p. 2). Yet in this condition of bondage, in their shared experience of misery, pain and wretchedness, they became increasingly aware of their sorry plight, and learned to yearn for freedom (Genovese, p. 114). The first stirrings of rebelliousness began to beat in their hearts yearning for a life free from bondage and servitude. In this shared experience, the slave community developed a culture distinctly its own, reverberating with the influence of their African origins infused with their newfound material conditions in American soil. It is the experience of a particular slave girl by the name of Linda Brent which we shall explore in detail. Jacobsââ¬â¢ work, Incidents in the Life of a Slave Girl, utilizes the literary form of the slave narrative, a form of autobiography with a unique structure and distinctive themes tracing the narratorââ¬â¢s path form slavery to freedom. It traces the narratorââ¬â¢s journey from poverty to freedom as her determination to overcome societal and self-imposed limitations leads her on to prevail despite her harrowing circumstances. It is quite a moving and inspiring tale, and the author succeeds in painting a deft picture of the real-life suffering of men and women in bondage who were born, raised and died as slaves at the hands of their masters in the sprawling plantations of the South. Though the slave narrative is recognized as a powerful literary form with obvious merits as a tool for anti-slavery and human rights causes as it compellingly illustrates how individuals could rise above the depths of their despair and overcome seemingly impossible odds, it too has its flaws as a resource for fully understanding the complex institution of slavery. At one point, it perpetuates the myth that individuals can overcome established social structures and societal features disadvantageous to marginalized groups, e. g. racism against colored peoples, through sheer determination, will power, a never-say-die attitude, and a perpetually rosy outlook in life that things could only get better. In actual truth, for most slaves the matter of winning oneââ¬â¢s freedom is a whole lot more complicated than what most narratives seem to suggest in their ââ¬Å"success stories. â⬠The abolition of slavery was not simply a case of dissolving a centuries-old institution due to the influence of writers who decided to go public with their personal accounts of the evils of slavery. In a way, the slave narrative is deceptive in its promise of deliverance (giving a sense of false hope) to blacks while reinforcing the notions of a superiority complex of whites over other colored peoples as they could always say that those who fail to break down societyââ¬â¢s barriers, e. g. racial boundaries and the gap between rich and poor, to achieve success have only themselves to blame for their failures. It thus becomes a case of ââ¬Å"If others canââ¬â¢t do it, why canââ¬â¢t I? â⬠Yet the situation is inherently more complex than this. In analyzing slavery one has to consider the larger macro factors ââ¬â economics, political institutions, cultural norms, ideology, etc. ââ¬â as shaping the micro ones. That is, in the case of the slave narrative one needs to situate the personal accounts in the larger social context of the period, the interplay between the personal and the historical, personal troubles and public issues, the intertwining of biography and history. Moreover, one could also apply a feminist reading in Jacobsââ¬â¢ text for as Linda laments the birth of her daughter Ellen, she remarks how ââ¬Å"â⬠¦slavery is terrible for men; but it is far more terrible for womenâ⬠(Jacobs, p. ). Incidents in the Life of a Slave Girl illustrates how slavery proved to be a more wretched state for women who had to endure the same dehumanizing cruelty and brutality inflicted on men, as well as the torment of sexual abuse at the hands of their male masters and the anguish of being taken away from their children. Their pain and degradation were further compounded as they suffered being used as vessels of lust for their masters, breeding bastards to add to their masterââ¬â¢s stock but denied a motherââ¬â¢s right to care for her children. The children born from such unions, Linda chillingly points out, was more often than not sold to protect the honor of the slave ownerââ¬â¢s wife faced with the undeniable living testimony to her husbandââ¬â¢s lust. Addressing the issue of human bondage from a womanââ¬â¢s perspective, Jacobs attempts to get through to her readers, particularly the women of the Northern states, to make them aware of their responsibility to make their voices heard in protesting against slavery for their silence would be in support of the perpetuation of slavery as an institution. Her tale emphasizes the struggle of a particular woman (herself) to protect her family, in the process learning to fight for her freedom to be an independent individual in control of her own life, and enticing her own family and community to join in the struggle for the emancipation and liberation of slaves. All said, in its own way Jacobsââ¬â¢ work has significantly contributed to the success of the movement to abolish slavery in the United States of America, and for that her efforts have been well-rewarded with the renewed scholarly interest in her work. Sucrose Synthase Key Enzyme In Sucrose Metabolism Biology Essay Sucrose synthase is a key enzyme in sucrose metabolism. Sucrose metabolism is required by the plant to form carbon required for various processes in the plant such as respiration, starch and cell wall formation. The enzyme is encoded by a small multigene family where most plants have at least two isoforms of the enzyme. The kinetics of sucrose synthase show that different Km values and ratios of sucrose breakdown exist for the enzyme. The methods of extracting, assaying and purifying the enzyme are shown in the enzyme characteristics. Factors such as pH, addition of different buffers, metal ions, fungal volatiles as well as environmental factors such as anoxia have all been shown to affect sucrose synthase activity. The enzymes protein sequences have been phylogenetically divided up into six main groups using clustalw. Sucrose synthase is normally present in the cytoplasm but the availability of sucrose in the chloroplast and its ability to use ADP as a substrate would indicate that the enzyme may be able to act in the chloroplast as well as the cytoplasm. Sucrose synthase is an important enzyme in sucrose metabolism in plants cells. (Persia et al., 2008) The main route of entry of carbon from sucrose is commonly known to be sucrose synthase. (Bieniawska et al., 2007) This carbon is used for respiration and in the synthesis of cell wall polymers and starch. (Persia et al., 2008) The main form of reduced carbon in plants is sucrose. It is used to support growth and synthesis of reserve materials e.g. starch in heterotrophic sink tissues. (Matic et al., 2004) The UDP-glucose supplied by sucrose synthase is used for cell wall biosynthesis while working with the cellulose synthase complex. (Baud, Vaultier and Rochat, 2004) In most fruit tissues, an increase in sucrose synthase activity is alongside with sucrose accumulation. This would suggest that sucrose synthase plays a physiologically important role. (Islam, Matsui and Yoshida, 1996) Carbohydrates are transported from photosynthetic source tissues to sink tissues in the form of sucrose . The consequent cleavage of sucrose in the sink tissues is the first step for its use in various metabolic pathways. The sugar is cleaved in vivo by either sucrose synthase (Sus) or by invertase. Invertase catalyses an irreversible reaction where sucrose is cleaved into glucose and fructose (Matic et al., 2004) while sucrose synthase catalyses the reversible conversion of sucrose and uridine-diphosphate (UDP) into uridine-diphosphoglucose and fructose. (Hirose, Scofield and Terao, 2008) (Hardin and Huber, 2004) These enzymes play a crucial role in plant growth and development. (Abid et al., 2009) Sucrose Synthase is cytosolic (Ã
ebkovà ¡ et al., 1995) and has been characterized in many different plant species such as maize (Hardin and Huber, 2004), rice (Odegard, Liu and Lumen, 1996) and sugarcane (Schà ¤fer, Rohwer and Botha (2005)). Its activity has been studied in many plant organs such as roots, leaves and seeds. (Ã
ebkovà ¡ et al., 1995) For trees, cellulose biosynthesis is a highly regulated process in which carbon is permanently placed in their primary and secondary cell walls. Sucrose is the main carbon source for cellulose synthesis. The stem is made up of extremely active sink cells which utilise sucrose for cellulose synthesis. Sucrose synthase is the main sucrolytic enzyme in these cells that catalyzes the reversible conversion of sucrose into fructose and UDP-glucose which is needed for cellulose biosynthesis. (Joshi, Bhandari and Ranjan, 2004) It also plays an important role providing adequate sugar supply during anoxic stress. It has been shown that during anoxic germination of rice, sucrose synthase activity was enhanced whereas the activity of invertase was depressed. This would indicate that sucrose synthase is the enzyme predominantly responsible for sucrose breakdown during anoxia. (Joshi, Bhandari and Ranjan, 2004) Fig 1: Diagram of the cleavage and synthesis reaction of sucrose synthase (Rà ¶mer et al., 2004) Different isoforms of the gene are present in most plants. In the case of maize, two non-allelic genes were discovered for sucrose synthase but more investigation lead to the discovery of a third. At least three genes for sucrose synthase have been discovered in rice where the genes show differences in expression between tissues. RSus1 is expressed in root phloem while RSus2 is expressed in leaf phloem. (Schà ¤fer, Rohwer and Botha, 2005) When examining the different isoforms at an amino acid level it is appears that there is less homology between different sucrose synthase genes in a species than when the gene is compared to its corresponding gene in another species. In the case of maize, there is 75% homology between the SS1 gene and SS2 gene of maize but there is 90% homology between rice RSus1 and maize SS2 genes. In sugarcane, the SS1 gene is 97% identical at the amino acid level to maize SS1 gene. (Lingle and Dyer, 2001) Nolte and Koch (1993) undertook a study to determine whether sucrose synthase was localized to certain part of the vascular strand. It is well known that sucrose synthase is present in vascular bundles for example in transgenic tobacco plants phloem specific expression of a maize sucrose synthase gene has been observed. Their study, using immunohistochemistry, found that sucrose synthase was restricted to the cytoplasm of companion cells of the phloem and did not appear to be present in other organelles of the plant. (Nolte and Koch, 1993) The molecular mass of sucrose synthase can be determined by gel filtration. Sucrose synthase elutes from the column with a Kav value of 0.17844 which when using a calibration curve correlates to a molecular mass of 362kDa. Using SDS-PAGE gradient gel the molecular mass of each subunit can be estimated at 92kDa. This can conclude that sucrose synthase is a tetrameric enzyme with a molecular mass of 360kDa and four identical subunits of 90kDa. (Hardin and Huber, 2004) (Elling and Kula, 1993) It can associate with membranes and the actin cytoskeleton where its activity is known to be involved with cellulose synthesis. It does this by channelling uridine-diphosglucose to the growing glucan chain by the enzyme cellulose synthase. (Hardin and Huber, 2004) Analysis of Sucrose Synthase Gene Family: From the results of species examined to date, it is shown that sucrose synthase is encoded by a small multigene family. (Bieniawska et al, 2007) Most species of plants have at least two isoforms of sucrose synthase. These isoforms usually have comparable biochemical properties and highly homologous amino acid sequences. (Wen et al., 2010) Further analysis of transgenic and mutant crop plants show certain isoforms of sucrose synthase have specific functions in the plant. The rug4 mutation of pea removes the SUS1 isoform but has no effect on SUS2 or SUS3. This would indicate that these two isoforms are not able to make up for the loss of SUS1 in the seed or root nodule. It is clear that the loss of different isoforms affect the plant in certain ways. Loss of the SH1 isoform in maize has different outcomes from the loss of SUS1 isoform. SH1 is required for normal cell wall formation during endosperm development while both isoforms are needed for wild-type rates of starch synthesis. Why different isoforms have different functions is unclear. The same functions can be carried out in the cell by different isoforms but can occur in distinct cell types, developmental periods or environmental conditions. It is likely that different isoforms could have non-overlapping, particular functions in the same cell. (Bieniawska et al., 2007) It is difficult to decide on the precise roles of the genes in sucrose synthase gene family when there is not enough information in existence. Although there is some information available on some of the isoforms and theyre functions in the plant, no analysis of the functions of the gene family has been carried out. The model plant Arabidopsis is ideal for carrying out such an analysis. Six sucrose synthase genes are in the Arabidopsis genome. Based on comparisons of the amino acid sequences the isoforms they encode can be divided into three distinct pair groups. The isoforms SUS1 and SUS4 are 89% identical to each other but have less than 68% similar amino acid sequences to other isoforms. Similarly, SUS2 and SUS3 are 74% identical to other isoforms and are 67% less identical to the other forms of enzyme. SUS5 and SUS6 are 585 identical to each other but have less 48% similarity to the other isoforms. When examining other dicotyledonous species it appears that at least two of the thr ee pairs of isoforms are present. When phylogenetic analysis was carried out, it showed that the isoforms AtSUS1 and AtSUS4 are related to pairs of isoforms from pea (Fabacae), carrot (Umbelliferae) and potato (Solanacae). A pair of isoforms from Craterostigma plantagineum (Scrophulariacae) is closely related to the pair of isoforms AtSUS2 and AtSUS3 in the Arabidopsis. The pair AtSUS5 and AtSUS6 is related strongly to a pair of genes from rice. This evidence shows that it is unlikely that the three pairs of isoforms in Arabidopsis are as a result of gene duplication events. It is possible that each isoform has an exact function preserved in a wide range of plants. The members of Arabidopsis gene family are strongly differentially expressed in different organs of the plant through its development and in response to external stimuli e.g. environmental stress. This is seen in gene families of other plants studied. (Bieniawska et al., 2007) Fruit quality is determined by the type and quality of sugars present. A study of the sucrose synthase-encoding gene from the muskmelon fruit was carried out to evaluate how to genetically improve the quality of the fruit. This is done by finding the sugar components in fruit, to identify the enzymes involved in sugar metabolism and distinguish the relationship between sugar accumulation and the activities of related enzymes. It is thought that sucrose synthase is the enzyme involved in metabolising sucrose in developing muskmelon fruit. To examine this, a full length cDNA strand encoding sucrose synthase was extracted from a muskmelon fruit by RT-PCR and RACE and identified as CmSS1. Real time PCR analysis showed that CmSS1 expression changed in among different tissues of the plant e.g. root, stem, leaf. It showed that the mRNA levels are highest in the root and lowest in mature fruit. Fig 2: The patterns of CmSS1 transcript abundance in the different tissues of the muskmelon plant. These results were found using quantitative real-time PCR analysis of total RNA prepared from the root, stem, leaf, flower and mature fruit of muskmelon. During fruit development and ripening it was shown that CmSS1 mRNA was at its maximum level at five days after pollination and decreased steadily during fruit development until it reached its minimum level of maturity. This was discovered using again real-time RT-PCR analysis of mesocarp tissues from five days of pollination to ripening. Fig 3: This graph depicts the patterns of CmSS1 transcript abundance in developing muskmelon fruits found by using quantitative real-time PCR analysis of total RNA prepared from muskmelon. (Wen et al., 2010) The sugar content and SS activity were analysed to show the functions of CmSS1 in regulating fruit quality. It showed that very low concentrations of sucrose are present in young and unripe muskmelons. Between 20 and 30 days after pollination there is a massive rise in the amount of sucrose in the fruit. Sucrose synthase activity increased in the direction of sucrose synthesis and decreased in the direction of sucrose cleavage through fruit development. (Wen et al., 2010) Enzyme Kinetics of Sucrose Synthase: An investigation was carried out by Schà ¶fer et al. to the find the properties of three sucrose synthase isoforms present in sugarcane. Kinetic analysis indicated that the three sucrose synthase genes in sugarcane are different isoforms, with major differences in Km values and the ratios of sucrose breakdown synthesis. The kinetic characteristics of the SuSyA and SuSyB isoforms, both expressed in the leaf roll, differ greatly. It was found that SuSyA has almost three times higher affinity for sucrose than the SuSyB isoform whereas SuSyB has a much greater affinity for UDP than SuSyA. Based on the differences in their kinetic properties it can be concluded that SuSyB and SuSyC are different isoforms of sucrose synthase. SuSyC has roughly ten times higher affinity for UDP compared to the other two isoforms. (Schà ¤fer et al., 2005) Fig 5: The graph shows the Lineweaver-Burk plot of 1/v against 1/S for the isoforms SuSys A, B and c where UDP was the variable substrate. The concentration of sucrose was kept constant at 320nM. The Km values were determined from the non-linear fit of the data to the Michaelis-Menten equation. (Schà ¤fer et al., 2005) When examining sucrose synthase in soybean nodules Morell and Copeland (1985) found the kinetic constants of UDP, UDPglucose, sucrose and fructose by fitting the data to the following two equations: 1. v = VA/KiaKh + KhA + KhB + AB 2. v = VA/Ka + A + A/Ki The kinetic constants for ADP, CDP and ADPglucose were found using non linear regression analysis of initial velocity data. (Schà ¤fer et al., 2005) Characteristics of Sucrose Synthase: Extraction of Protein: The method for extracting protein from the leaves of maize (Zea mays), rice (Oryza sativa) and tobacco was done as follows: 1-3g of leaves was ground in liquid nitrogen and the powder was mixed in the ratio 1:2 with extraction buffer. The buffer was made up of 0.1M tris-HCl, pH 8, 10mM DTT and 1% polyvinylpolypyrrolidone. The samples were then incubated on ice for 15 minutes and then centrifuged at 1,000g for 10 minutes at 4oC. The pellet was then removed and the supernatant was re centrifuged at 100,000g for one hour at 4oC. After this final centrifugation, the pellet and supernatant which contained the soluble proteins was resuspended in sample buffer for electrophoresis. (Persia et al., 2008) When extracting protein from rice seeds, a similar procedure is followed. Seeds weighing roughly 50-100mg at various stages of growth were homogenized in 400à µl of extraction buffer and kept at 4oC. The buffer was made up of 50mM Tris-HCl, pH7.5, 1.0mM DTT, 1.0mM EDTA and 2mM PMSF. Ammonium sulphate fractions (30-50% w/v) were precipitated and then resuspended in dialysis buffer made up of 50mM Tris-HCl, pH 8.0, 5mM MgSO4, 5mM 2-mercaptoethanol. This was then dialyzed overnight at 4oC. (Odegard, Liu and De Lumen., 1996) The method for extracting protein from tobacco pollen tubes is slightly different to those mentioned previously. The pollen first was slowly thawed from storage at -20oC and hydrated in a humid chamber overnight. It was then germinated in BK medium and allowed to germinate at 25oC for three hours. After this period had elapsed, the pollen was collected by centrifugation at 1,000g for 5 minutes at 25oC. It was then washed twice with BRB25 buffer which is made up of 25mM HEPES, pH 7.5, 2mM EGTA and 2mM MgCl2 and 15% Suc. After washing, the pollen was resuspended in lysis buffer and lysed on ice using a motor-driven Potter-Elvehk-jem homogenizer. The lysis buffer used was made up of BRB25 buffer along with 2mM dithiothreitol, 1mM phenylmethylsulfonyl fluo ride (PMSF), 10à µL/mL protease inhibitors, 1mM NaN3 and 10% mannitol. After lysis was carried out, the samples were centrifuged at 1,000g for 10 minutes at 4oC. The supernatant was centrifuged again at 4oC for 45 minutes at 100,000g over a 20% (w/v) Suc cushion. The supernatant was then collected as it contained the soluble protein fraction. (Persia et al., 2008) Enzyme Assays: After extracting protein, the sucrose synthase activity in sugarbeets was found using a spectrophotometric end point assay. The activity of the enzyme was monitored as fructose formed at 35oC. This was carried out in a solution that contained 250mM sucrose, 2mM UDP and 100mM MES. The control was carried out by assaying for activity in the absence of UDP. The total protein concentration was determined using the Bradford method where bovine serum albumin was the standard. (Klotz and Haagenson., 2008) When assaying for protein from rice, the Bradford method was followed to determine protein concentration as was done in Klotz et al. 40mg of protein was used per assay. The assay was carried out in 20mM MES pH 6.4, 200mM sucrose and 4mM UDP for 15 minutes at 30oC. The reaction was stopped by boiling for 2 minutes and the fructose levels were measured. The control tubes did not contain UDP. (Odegard, Liu and De Lumen., 1996) When examining the effect of sucrose synthase on carbon partitioni ng a similar method was followed for assaying the protein. Sucrose synthase was assayed in the direction of sucrose breakdown using 50à µl poplar plant extract. The tetrazolium blue assay was followed to determine the amount of free fructose. As in previously mentioned assays, the absence of UDP in the assay acted as a control. The total protein content was found by employing the Bradford (Bio-Rad) protein assay. (Coleman, Yan and Mansfield., 2009) A similar method was followed for carrying out an assay for the enzyme on tomato tissue. The reaction mixtures contained 50mM Hepes-NaOH buffer, 15mM MgCl2, 25mM fructose and 25mM UDP glucose. This was incubated at 37oC for 30 minutes and was terminated with the addition of 70à µl of 30% KOH. The enzyme blanks were terminated with the addition of KOH at 0 minutes. The tubes were then kept at 100oC for 10 minutes to destroy any fructose. The soluble protein content was determined using the Lowry method whereby bovine serum albumin was th e standard. (Islam, Matsui and Yoshida., 1996) Alkaline copper solution is added to each tube and allowed to stand at room temperature for roughly 30 minutes. Dilute folate reagent is then added to each tube rapidly and after 30 minutes the absorbance is read at 750nm. (Lowry et al., 1951) The results were measured as à µmole of sucrose per minute per mg protein. (Islam et al., 1996) When assaying for sucrose synthase in the cleavage direction Rà ¶mer et al used recombinant SuSy1 gene from potato. In a volume of 100à µl HEPES buffer with a concentration of 200mM and pH 7.6 recombinant sucrose synthase was incubated along with 2mM UDP and 500mM sucrose for ten minutes at 30oC. HPLC analysis was used for the formulation of UDP-glucose. The Bradford assay was used to determine protein concentrations as was carried out by Klotz et al and Coleman et al. The activity of the enzyme was also tested with the nucleoside diphosphates dTDP, CDP, ADP and GDP at 2mM. For assaying recombinant e nzyme in the synthesis direction a similar method was followed as when assaying for standard enzyme. Recombinant sucrose synthase was incubated in a total volume of 100à µl HEPES buffer where this time the pH was 8.0 and the concentration was as in cleavage direction of 200mM. 1mM UDP-Glc and 20mM D-fructose was also added to the mixture and it was incubated for five minutes at 30oC. The reaction was heated to 95oC for five minutes and HPLC analysis was used to establish the formation of UDP. The sucrose synthase activity was also tested using dTDP-Glc, CDP-Glc and ADP-Glc. (Rà ¶mer et al., 2004) Purification of Protein: After extraction of the protein from the crude extract, purification can be carried out. This can be done in a number of ways such as Batch adsorption with Sephadex A50, Anion exchange chromatography and Gelfiltration. SDS-PAGE can be carried out after purification to check the purity of the protein sample. The Sephadex A50 gel is loaded into a glass funnel and washed twice with deionised water. The gel was then washed twice with 300ml standard buffer. The protein sample was loaded to the gel and slowly sucked through the gel for 30 minutes. The gel bed was then washed with 300ml standard buffer and then with 300ml standard buffer containing 100mM KCl. The last washing step contained 300mM KCl. 200ml of the first salt preparation was concentrated to 40-50ml by using a cross-flow ultrafiltration module with YM 30 ultrafiltration membrane that had been pretreated with 55 PEG 4000 solution. This was done to prevent the enzyme sticking to the membrane. In anion exchange chromatography a Sepharose Q column was first equilibrated with 300ml Hepes buffer. This was made up of 200mM pH 8 with 50mM KCl. 70-80mg of protein sample was loaded and the elution was started using two different salt gradients. To prevent enzyme inactivation after elution all the fractions were titrated back to pH 7.2. All fractions that contained enzyme activity were pooled and concentrated by using ultrafiltration. Gelfiltration experiments are carried out on a prepacked HiLoad 16/60 Superdex 200 prep grade column that was connected to FPLC equipment. Four samples containing 2mg of protein were loaded and eluted with a flow rate of 1 ml min-1. The fractions were then pooled and stored at -20oC in 500à µl aliquots. (Elling and Kula., 1993) To determine the purity of the protein, SDS-PAGE is carried out. This is done by loading 100à µg of protein samples to a 125 SDS-polyacrylamide slab gel that was overlaid with stacking gel. The electrophoresis was carried out at 4oC and at 40V for 16 hours a nd followed by 200V for one hour. Coomassie blue R 250 was used to stain the gel followed by destaining. ebkovà ¡ et al (1995) stated that sucrose synthase has two different pHs for optimal activity. In the cleavage direction it was found that most enzyme activity was observed between pH 6.0 and 8.5 at temperatures between 50 to 55oC. ebkovà ¡ et al., 1995) This would correlate with the findings of Morell and Copeland (1985) who found that optimal activity of the enzyme in soybean was at pH 6 in the cleavage direction and at a pH of 9.5, sucrose synthase activity in the synthesis direction was at its highest. It was also found that at a pH of 7.5 the cleavage and synthesis activities were their highest. (Morrell and Copeland., 1985) Elling and Kula (1995) examined the effect of buffers TES-NaOH, MOPS-NaOH, TEA-NaOH and Tris-HCl on the pH optimum of sucrose synthase activity. These were determined using UDP and TDP as substrates for the reaction. They found that the e nzyme had its highest activity in Hepes-NaOH buffer. When MOPS-NaOH and TES-NaOH buffer was used, only 60-80% activity was noted. (Elling and Kula 1995) It was also found that the velocity of the reaction could be increased by increasing the temperature where optimal activity was seen between 50 and 60oC. Xu at al (1989) reported that potato and bean are also able to withstand these high temperatures. However once the temperature goes above 60oC enzyme activity starts to decreased rapidly and was destroyed once the temperature reached 70oC. (Xu et al., 1989) The cleavage of sucrose by the sucrose synthase enzyme was investigated to find the rate of cleavage reaction using different nucleosidediphosphates as cosubstrates. They found that the rate of reaction was UDP>TDP>ADP>CDP>GDP. Echt and Chourey (1985) found similar results when examining nucleotide specificity. They found that substrate specificity for SS1 and SS2 were UDP>TDP>ADP>CDP>UTP where each substrate was at a concentrat ion of 4mM. (Echt and Chourey 1985) Low levels of heavy metal ions such as mercurate inhibited cleavage activity of the enzyme. The activity was lessened slightly (10%) by the presence of 1mM Mn2+ and Mg2+ ions with UDP. The enzyme is completely inactivated in the presence of 1mM Cu2+ or Fe2+. (Elling and Kula., 1993) A recent study was undertaken to examine the effects of volatile emissions on carbohydrate metabolism. Studies on this area have taken place before but it is usually examining the results of physical contact between the host plant and the microbe. No work has taken place until now on the effect on the plant in the absence of physical contact. Many microbes such as Pseudomonas spp, Strepomyces spp, Penicillin spp and a selection of truffles produce ethylene. This gaseous plant hormone plays an important role in many aspects of plant growth and development such as seed germination, root hair initiation, fruit ripening and starch accumulation. In the work of Ezquer et al (2010), the possible effects of volatiles released from gram-negative bacteria, gram-positive bacteria and fungi on starch metabolism was studied. The results showed that the volatile compounds released by microbes promoted high levels of starch accumulation in mono- and dicotyledonous plants. It also revealed fungal vo latiles (FVs) promoted massive changes in expression of genes involved in many important processes in plant such as metabolism of carbohydrates, amino acids, sulphur and lipids, energy production, protein translation and stability, cell wall biosynthesis and photosynthesis. However no changes were noted in the expression in some of the genes that coded for proteins involved in starch and sucrose metabolism such as plastidial hexokinase, plastidial phosphoglucose isomerase, plastidial adenylate kinase, alkaline invertase and UDPglucose (UDPG) pyrophosphorylase. It was found in the study that FVs strongly upregulate the expression of Sucrose Synthase in potato leaves. The plants were cultured in the presence and absence of FVs emitted by A. Alternata. This caused a massive enhancement of expression of Sus4 isoform. A 29.4- and 31.63-fold increase was observed in expression when the plants were cultured in the presence and absence of sucrose. This isoform of the enzyme controls the accumulation of ADPG, UDPG and starch in potato source leaves and tubers. Analyses of the intracellular amounts of starch and nucleotide-sugars in the leaves of the plant show a positive correlation between patterns of enzyme activity and starch, UDPG and ADPG amounts. This was noted when the leaves were cultured in the presence and absence of FVs. Western blot analyses and quantitative RT-PCR confirmed also the increase in expression. (Xu et al., 1989) Environmental Factors affecting Sucrose Synthase Activity: Anoxia: Waterlogging is where oxygen supply is blocked to root leading a severe decrease in the amount of oxygen available to the plant. This leads to inhibition of root respiration that causes a major decline in energy of root cells affecting vital metabolic processes of the plant. This is restriction of oxygen supply is known as anoxia. The presence of glucose in an anoxic incubation medium drastically decreases meristem death and studies have shown that sucrose synthase is the enzyme mainly responsible for sucrose breakdown under anoxia. (Kumutha et al., 2008) The increase in glycolytic demands caused by these demands is the cause of increased sucrose synthase expression. This has been demonstrated in many plant species e.g. sucrose synthase gene is induced in wheat and in rice when oxygen levels are low. (Ricard et al., 1998) Harada et al (2005) also found an increase in sucrose synthase activity in pondweed turins while under anoxia. (Harada et al., 2005) Klotz and Haagenson (2008) foun d that sugarbeet contained two genes for sucrose synthase activity-SBSS1 and SBSS2. They demonstrated that anaerobic conditions caused a large increase in the transcription levels of SBSS1 and a quick increase and succeeding decline in SBSS2 transcription levels. However this did not correlate with a significant increase in sucrose synthase enzyme activity. A 23% increase in sucrose synthase activity was noted after initiation of anaerobic conditions but otherwise the activity of the enzyme did not differ greatly to that of the controls. Balance Between Sense and Sensibility in Jane Austen's Northanger Abbey Throughout her novel, Northanger Abbey, Jane Austen integrates parody with characterization to emphasize the necessity of a balance between sense and sensibility while reflecting a theme of the initiation of a young woman into the complexities of adult social life. This novel can be traced back as one of Jane Austen's earliest works. It was written in 1798, but not published until 1818, and is an excellent example of what Austen believed a novel should not be. In the work Jane Austen's Novels Social Change and Literary Form, Julia Prewitt Brown states "The evident purpose of Northanger Abbey is to burlesque the popular fiction of her day, to carry its conventions and assumptions to an absurd extravagance" (50.) To achieve her purpose Austen uses parody to portray a comic version of a gothic novel while presenting false emotions of romanticism and concentrating on pure human beings and their mutual reactions. "The writer of the pure novel sets out to delight us not by prodigality of invention, the creation of a large gallery of characters, the alternation of a large number of contrasted scenes, but by attention to the formal qualities of composition, to design, to the subordination of the parts to the whole, the whole being the exploration of the relations between his characters or of their relations to a central situation or theme." (Allen, pp114.) In Northanger Abbey, Austen intended to reflect a contrast between a normal, healthy-natured girl and the romantic heroines of fiction thorough the use of characterization. By portraying the main character, Catherine Moorland, as a girl slightly affected with romantic notions, Jane Austen exhibits the co... ...ne show his sensibility. His imagination and creativity motivate him to read Gothic romances and to indulge in the effects that his inventive tales produce. His decision to marry Catherine is motivated by feelings of love that further exemplifies his sensibility. Throughout the novel the readers see an excellent display of Henry's ability to maintain equilibrium between the two qualities. He passes his knowledge onto Catherine to help her to become a better person. At the end of the novel it is apparent that Henry has taught the keys of his success to Catherine. NatWest Bank is a member of the Royal Bank of Scotland Group (RBS Group). In 1968 National Provincial Bank (est.1833) and Westminster Bank (est.1836), fused as National Westminster Bank. This is a necessary reward for the costs of R&D, and helps to encourage innovation. Nevertheless, the internal return to innovation may still be modest relative to the industry-wide benefits that arise once the innovation can be replicated by other firms. â⬠¢ Inability to diversify risk: While larger companies may have a large portfolio of innovative activity, innovation in smaller firms may be concentrated on a single product or process, and the cost of undertaking the necessary R&D may be very large relative to the overall financial assets of the company. Accordingly, where SMEs are risk-averse, they will be discouraged from investing in innovative activities with uncertain returns. Assembling the enterprise confront will rely on the innovation and creativity of entrepreneurs themselves, but the Government must also provide the right cond itions and the necessary support for successful enterprise. Costs would also be involved in the provision of such items as HR policies, for example the sunk costs associated with setting up employee benefit programmes, such as pension plans, stock options, bonus schemes, car schemes, health care etc. The same tactic has been adopted and this turns up at an total expenditure of à £ [255] m. This is probably a conservative estimate. Over the period 1996 to 2000 NatWest incurred expenses of around à £ [13] bn in respect of its Retail Transformation programme, which was only a modification of part of its existing infrastructure rather than creation of a totally innovative one. (RBSG, 2002) NatWest depends entirely upon the qualities of innovation, and suffer more acutely from the rapidity of transform. Whether in presentation, writing, the visual arts or advertising, the progress of new ââ¬Ëproductââ¬â¢ is at the heart of all doings. Without this self- generated action, there is not anything to put up for sale. The sector therefore has a specific and challenging manufacturing process or value chain which, at each stage, demands different talents. Since about 1980, with the decline of heavy industries and growth of a service economy, like other clearing banks, BoS was obliged to expand further into small business lending and new retail products (e.g. residential mortgages, financial services, credit cards, etc.). Competition was exaggerated by banking deregulation, which led to the growth of a secondary banking sector, the transformation of many building societies (including Halifax) into banks, and new amalgamations into ever-larger banks (as cited in Leyshon and Thrift 1993). During this period there was increasing concern in the Scottish banking community about the potential for takeovers to erode a distinctive Scottish banking sector (as cited in Saville 1996: 717-40), which had been partly maintained by an historical ââ¬Ëgentlemanââ¬â¢s agreementââ¬â¢ between the English and Scottish banks to limit their presences in each otherââ¬â¢s markets. In September 1999, BoS surprised The City of London by making a à £20.85 billion bid to take over NatWest Bank, in effect striking first in an environment where further bank mergers seemed inevitable. Many in the financial press were quick to point out that under these new terms, BoS and other Scottish banks could not expect to have the ir future ââ¬Ëindependenceââ¬â¢ protected. In late November the Royal Bank of Scotland, BoSââ¬â¢s main competitor in Scotland, made a successful counter bid of à £25.1 billion. The City now anticipated a takeover of BoS, compelling BoS to keep pace with the Royal Bank. BoS and Halifax began merger negotiations in April 2001, and the new banking group, HBOS, began trading on the stock market on 10 September, 2001. The Halifax was the dominant partner in the merger, being about twice the size of BoS in terms of number of employees and market value at the time of merger, and supplying key organizational leadership, including the CEO. The core rationale of the merger was that it brought together Halifaxââ¬â¢s substantial mortgage lending income and BoSââ¬â¢s expertise and placement in the corporate banking world to create new business opportunities for the merged organisation. It also saw the dislocation of BoSââ¬â¢s rather conventional, cautious and habitual banking ethos by the more modern, competitive and market driven ethos of the Halifax organisation. Thus the merger was experienced by staff as an encounter between two different corporate cultures, and two different national cultures, at the same time. By ââ¬Ëthe ideology of changeââ¬â¢ it specifies an ideological condition that corresponds closely with Therbornââ¬â¢s conception of modernity. With this phrase I want to dire ct attention not so much to a set of ideas and beliefs about change, but more to a disposition, or attitude toward change. I mean to suggest a normatively charged attitude in which established ways of doing things are devalued and innovation and change is positively valued in principle, regardless of the particulars of any given situation. It is, in a sense, a ââ¬Ëpresumption of guiltââ¬â¢ in regard to the old, and ââ¬Ëpresumption of innocenceââ¬â¢ in regard to the new. This attitude is deeply naturalised, so that the imperative and positive value of change is widely regarded as self-evident, and not easily questioned. Thus rather than the great social critiques and political programmes associated with the formation of modernity, I mean to invoke a routinized and normalized aspect of established modernityââ¬âseen in the way political parties and governments promote themselves through promises of reform, in the way corporations and public institutions are constantly restructuring in order to ââ¬Ëmodernizeââ¬â¢ and keep pace with their competitors, and in the way consumer-citizens in capitalist society come to expect scientific and technological advances that will increase knowledge and improve commodities and services, while fashions in popular culture rapidly replaces one another. The ideology of change is mundane, relentless common sense. Some problems of ââ¬Ëinnovation and creativityââ¬â¢ lay with the organization and its leadershi p, rather than personnel, the exhortation to embrace change was ultimately being translated into a message of personal moral reform. In this way the ideology of change, while meeting resistance, percolated down to beleaguered selves seeking some greater purchase on their state of affiars. (Hearn 2006) HRM Strategic Milestones in NatWest: During 1990ââ¬â91 County NatWest, an investment bank, asked all its business units, including its personnel department, to set up strategic milestones for a five-year period. Their performance was to be measured against those milestones at specified target dates. The requirement to produce strategic milestones as an input to the bankââ¬â¢s five-year marked an important watershed in defining the contribution of personnel to the business at a strategic level. It forced the department to reflect on the nature of that contribution. Senior management of the bank duly authorised 18 separate strategic milestones. The milestones were consistent one with the other, and overall addressed issues that consultation within business units and across the three personnel teams had shown to be critical to business success. Each milestone was assigned to a designated individual and was incorporated into his or her own targets of performance. Quarterly reviews on progress, involving the whole department, were subsequently held to ensure that the milestones were on target. The operational tasks that a centralised T&D planning process involves seek to ensure that: T&D considerations are taken fully into account when business strategy is formulated. They form part of a human resource plan within the wider business plan. At business unit/divisional level there are policies to ensure people are trained and developed in line with the needs of the business. At the individual level T&D is an integrated part of daily routine and procedures, helping people to achieve performance standards and behavioral objectives and building up the kind of workforce needed in terms of productivity, quality and flexibility. T&D staff operates a collaborative approach to planning at every stage in order to ensure a high level of buy-in from line management and other stakeholders. Through the darkness, a staccato burst from the nose of the chopper sent a torrent of bullets chewing across the Goya's fiberglass deck, slashing a line across the stern. Rachel dove for cover too late and felt the searing slash of a bullet graze her arm. She hit the ground hard, then rolled, scrambling to get behind the bulbous transparent dome of the Triton submersible. A thundering of rotors exploded overhead as the chopper swooped past the ship. The noise evaporated with an eerie hiss as the chopper rocketed out over the ocean and began a wide bank for a second pass. Lying trembling on the deck, Rachel held her arm and looked back at Tolland and Corky. Apparently having lunged to cover behind a storage structure, the two men were now staggering to their feet, their eyes scanning the skies in terror. Rachel pulled herself to her knees. The entire world suddenly seemed to be moving in slow motion. Crouched behind the transparent curvature of the Triton sub, Rachel looked in panic toward their only means of escape-the Coast Guard helicopter. Xavia was already climbing into the chopper's cabin, frantically waving for everyone to get aboard. Rachel could see the pilot lunging into the cockpit, wildly throwing switches and levers. The blades began to turnâ⬠¦ ever so slowly. Too slowly. Hurry! Rachel felt herself standing now, preparing to run, wondering if she could make it across the deck before the attackers made another pass. Behind her, she heard Corky and Tolland dashing toward her and the waiting helicopter. Yes! Hurry! Then she saw it. A hundred yards out, up in the sky, materializing out of empty darkness, a pencil-thin beam of red light slanted across the night, searching the Goya's deck. Then, finding its mark, the beam came to a stop on the side of the waiting Coast Guard chopper. The image took only an instant to register. In that horrific moment, Rachel felt all the action on the deck of the Goya blur into a collage of shapes and sounds. Tolland and Corky dashing toward her-Xavia motioning wildly in the helicopter-the stark red laser slicing across the night sky. It was too late. Rachel spun back toward Corky and Tolland, who were running full speed now toward the helicopter. She lunged outward into their path, arms outstretched trying to stop them. The collision felt like a train wreck as the three of them crashed to the deck in a tangle of arms and legs. In the distance, a flash of white light appeared. Rachel watched in disbelief and horror as a perfectly straight line of exhaust fire followed the path of the laser beam directly toward the helicopter. When the Hellfire missile slammed into the fuselage, the helicopter exploded apart like a toy. The concussion wave of heat and noise thundered across the deck as flaming shrapnel rained down. The helicopter's flaming skeleton lurched backward on its shattered tail, teetered a moment, and then fell off the back of the ship, crashing into the ocean in a hissing cloud of steam. Rachel closed her eyes, unable to breathe. She could hear the flaming wreckage gurgling and sputtering as it sank, being dragged away from the Goya by the heavy currents. In the chaos, Michael Tolland's voice was yelling. Rachel felt his powerful hands trying to pull her to her feet. But she could not move. The Coast Guard pilot and Xavia are dead. We're next. 111 The weather on the Milne Ice Shelf had settled, and the habisphere was quiet. Even so, NASA administrator Lawrence Ekstrom had not even tried to sleep. He had spent the hours alone, pacing the dome, staring into the extraction pit, running his hands over the grooves in the giant charred rock. Finally, he'd made up his mind. Now he sat at the videophone in the habisphere's PSC tank and looked into the weary eyes of the President of the United States. Zach Herney was wearing a bathrobe and did not look at all amused. Ekstrom knew he would be significantly less amused when he learned what Ekstrom had to tell him. When Ekstrom finished talking, Herney had an uncomfortable look on his face-as if he thought he must still be too asleep to have understood correctly. ââ¬Å"Hold on,â⬠Herney said. ââ¬Å"We must have a bad connection. Did you just tell me that NASA intercepted this meteorite's coordinates from an emergency radio transmission-and then pretended that PODS found the meteorite?â⬠Ekstrom was silent, alone in the dark, willing his body to awake from this nightmare. The silence clearly did not sit well with the President. ââ¬Å"For Christ's sake, Larry, tell me this isn't true!â⬠Ekstrom's mouth went dry. ââ¬Å"The meteorite was found, Mr. President. That is all that's relevant here.â⬠ââ¬Å"I said tell me this is not true!â⬠The hush swelled to a dull roar in Ekstrom's ears. I had to tell him, Ekstrom told himself. It's going to get worse before it gets better. ââ¬Å"Mr. President, the PODS failure was killing you in the polls, sir. When we intercepted a radio transmission that mentioned a large meteorite lodged in the ice, we saw a chance to get back in the fight.â⬠Herney sounded stunned. ââ¬Å"By faking a PODS discovery?â⬠ââ¬Å"PODS was going to be up and running soon, but not soon enough for the election. The polls were slipping, and Sexton was slamming NASA, soâ⬠¦ ââ¬Å" ââ¬Å"Are you insane! You lied to me, Larry!â⬠ââ¬Å"The opportunity was staring us in the face, sir. I decided to take it. We intercepted the radio transmission of the Canadian who made the meteorite discovery. He died in a storm. Nobody else knew the meteorite was there. PODS was orbiting in the area. NASA needed a victory. We had the coordinates.â⬠ââ¬Å"Why are you telling me this now?â⬠ââ¬Å"I thought you should know.â⬠ââ¬Å"Do you know what Sexton would do with this information if he found out?â⬠Ekstrom preferred not to think about it. ââ¬Å"He'd tell the world that NASA and the White House lied to the American people! And you know what, he'd be right!â⬠ââ¬Å"You did not lie, sir, I did. And I will step down if-ââ¬Å" ââ¬Å"Larry, you're missing the point. I've tried to run this presidency on truth and decency! Goddamn it! Tonight was clean. Dignified. Now I find out I lied to the world?â⬠ââ¬Å"Only a small lie, sir.â⬠ââ¬Å"There's no such thing, Larry,â⬠Herney said, steaming. Ekstrom felt the tiny room closing in around him. There was so much more to tell the President, but Ekstrom could see it should wait until morning. ââ¬Å"I'm sorry to have woken you, sir. I just thought you should know.â⬠Across town, Sedgewick Sexton took another hit of cognac and paced his apartment with rising irritation. Where the hell is Gabrielle? 112 Gabrielle Ashe sat in the darkness at Senator Sexton's desk and gave his computer a despondent scowl. Invalid Password ââ¬â Access Denied She had tried several other passwords that seemed likely possibilities, but none had worked. After searching the office for any unlocked drawers or stray clues, Gabrielle had all but given up. She was about to leave when she spotted something odd, shimmering on Sexton's desk calendar. Someone had outlined the date of the election in a red, white, and blue glitter pen. Certainly not the senator. Gabrielle pulled the calendar closer. Emblazoned across the date was a frilly, glittering exclamation: POTUS! Sexton's ebullient secretary had apparently glitterpainted some more positive thinking for him for election day. The acronym POTUS was the U.S. Secret Service's code name for President of the United States. On election day, if all went well, Sexton would become the new POTUS. Preparing to leave, Gabrielle realigned the calendar on his desk and stood up. She paused suddenly, glancing back at the computer screen. Enter Password:_ She looked again at the calendar.

I will imagine myself a member of the Jewish council, describing the conditions of ghetto life and reflect on my role and relationships inside the ghetto. Ghettos were set up all over Poland and the Soviet Union, with some of the major ghettos Including Warsaw, Load, Love, Lubing, Krakow, and Bilabials. The German authorities were supposed to oversee the dally actively of the Jews Inside the ghettos. Instead, the Nazis appointed Jewish Councils or Juddered In each ghetto to implement Nazi policies. The Jewish Council served at the whim of the German authorities but also tried to be the voice for the Jews. In each ghetto the Jewish Council also distributed scarce resources, organized social life, set up charities, and tried to find ways to maintain some kind of human community (Genocide 1 15). As the book explains, They [Nazis] appointed recognized Jewish leaders-prominent people, businessmen, teachers, lawyers-to these boards and assigned them the task of carrying out German orders within the ghettos (Genocide 115). Living conditions Inside the Ghettos were absolutely horrendous with overcrowding, starvation, disease, little running water or electricity, lack of medical aid, and hard labor. Alternative ways to feed their females by smuggling In food or sending their children out to find food became prominent as the Jews sought any means possible to keep themselves fed. There was already famine at that time in the Ghetto, and the streets were littered with people dying of hunger (Images 137). Very little resistance was possible in most of the ghettos, as the Nazis kept a strict eye on everything. As we saw in the movie, The Pianist, a young boy was beaten to death limning through a small hole in the wall trying to bring food back into the ghetto (The Pianist). Jews were given small rations of food a week that could barely feed one person. Jews were made to contribute to the German war effort by working in German factories making uniforms, boots, bed linen, and underwear. The reason why working cards were so essential was because It was the only way the Jews could officially stay In the ghettos. They were given more of an opportunity to not be deported to the death camps by working. Jews who didnt have work cards frantically tried to get them. We saw that In the movie when Adrian Brood got his father a work card to save him. He believed that if he got everyone in his family a work card then teen would stay together ( plants). I nee wealthy Jews were Trace to live Walt poor Jews but they seemed to live a better life by buying what they wanted. Only the wealthy Jews could buy on the black market. The Pianist gave us an idea about the class divisions between the wealthy and poor Jews. Adrian Brood played a pianist, and in one scene he was playing in a cafeà © where the wealthy Jews ate. While the poor Jews were on the streets dying, starving, and dirty, the wealthy Jews were smoking, ringing, laughing, and listening to good music (The Pianist). The Jewish Councils relationship with the Nazis was an uncommon one. To a lot of the Jewish leaders it was a burden. They were powerless to help but yet they were forced to do what the Nazis wanted. In cases when members of the councils refused to cooperate, German officials dismissed them or had them shot and then replaced with more compliant men (Genocide 115). Some believed that cooperating with the Germans would help them survive longer but they too eventually were killed. There was a love hate relationship with their fellow Jews. Above them loomed German orders; below them spread the ever more desperate needs of the Jewish communi ties (Genocide 116). Some of the Jews resented the Jewish Council, saying they should have warned them or done more. They didnt like the Council collaborating with the Germans. The Jewish Councils tried to help their people, to maintain order, save lives, and to feed, clothe, and doctor the Jews in the ghettos (Genocide 116). There was hostility from their own people and in some ways it pushed them to behave the opposite of their goal. They tried to mediate and plead on the Jews behalf. Some Council members helped with the resistance and some believed it would doom the entire ghetto. An example of the Jewish Council in the ghetto is, New proclamations from the Juddered have been hung up which have caused panic among the Jews. The families of those working are no longer protected (Images 161). The Jewish Council formed its own Jewish Order Police. The Jewish police were also made to enforce order and deport Jews following the commands of the Germans. Like the Council, the Order Police were also disliked among the ghetto tenants. The purpose of the police was to prevent crime, supervise notation, and direct traffic. The Jewish police delivered to the Germans exactly the number of people needed, rounding them up any way they could, beating and kicking those who didnt want to go (Images 141). They collected ransom payments, stole valuables, fetched people for forced labor, forced payment of taxes, guarded the gates, and rounding up Jews for deportations. One of the problems with the police was evident in, A Cup of Tears, The Jewish police have been looting, breaking open flats, emptying cupboards, smashing crockery and destroying property, Just for the UN of it (Images 158). Henry Gordon proclaimed, It wasnt long before the Jewish police had an opportunity to profit even more from our misery (Images 141). There were a lot of problems and tension with the Jewish police. Ghetto police carried out the orders of the Nazis, assisting with deportations, punishment and oppression. Deportation required organization on a massive scale with the help of many people and organizations. In, The Pianist, we saw how Jews were rounded up in small areas waiting for the trains. Some of the Jews were questioning it and some didnt want to believe that they were being sent to their death. Obviously they never came back but no one really knew for sure what was happening (The Pianist). Every day children became orphaned, and many had to take care of even younger children. Orphans often lived on the streets, begging for bits of bread from others who had little or nothing to share. Many froze to death in the winter. They were broken full of despair. Simon tells us that, Little Eli in the Ghetto survived miraculously the many raids on the children, who were looked upon as non-working, useless mouths (Images 137). Women and children were usually the first to be deported because they werent as useful as the men. It looks like there is a policy to liquidate women and children (Images 156). The fear of keeping their wives and children fed and hiding them from the Germans consumed most men. Kristin Kern explains, My father was always looking for places to hide my little brother, Pale, and me because the Germans were intent on getting rid of all the Jewish children (Images 46). The way that families dealt with their fate and their families fate included giving up their children to save themselves, hiding, working, becoming part of the resistance, committing suicide, doing whatever you can to fed and clothe your family, and escape. Some people followed their families on the deportation trains even when they had work cards because they didnt want to leave heir family. I personally would hide, escape, and do anything to save my family and myself. Many Jews responded to the ghetto restrictions by forming Jewish resistance groups and engaging in illegal activities. Others believed that becoming part of the Jewish Council mediating between the Germans and Jews would help maintain order and save lives and some became servants to the Germans, with the power and cruelty of the Jewish Police. In the end, ghettos were used as a central step in the Nazi process of control, demutualization, and annihilation of Jews.

Another episode occurs in a different part of Los Angeles in which the cops Ryan and Hanson show racism by pulling over on Cameron an African American director together with his wife. The attack occurs without any apparent cause; Ryan goes a head to sexually molest the Christine the wife of Cameron in front of her husband who appears helpless at this moment. He justifies his actions by saying he was searching for hidden weapons in Christineââ¬â¢s cloths. The cops are supposed to provide security and ensure that victims of racial prejudice are defended according to the law. When the agents of security instead turn out to be the racists then it becomes trickier for the victims. It appears like the cops would rather attend to cases where fellow whites are victims rather than where the African Americans are the victims. The acts of the cop towards Christine are very dehumanizing considering that the cop was molesting her right in front of her husband. The scene at Cobaltââ¬â¢s house where a locksmith Mr. Michael Pena was called to change the lock also shows a racist act by Sandra. Sandra is in an argument with her husband Rick to change the locksmith. Being black, Sandra argues that the locksmith was a member of the gang that carjacked them. The locksmith overhears the argument and the negative comments directed to him. The locksmith leaves feeling insulted; this scene carries the prejudice that Sandra has towards the blacks and especially after being carjacked by the two young black men. The locksmith was indeed not a member of the gang but because of the prejudice that Sandra had towards the blacks, she generalizes the experience she had from the black thieves to all blacks. According to her, any black is a car thief and she does not want to interact with them in any way. The locksmith is apparently very sensitive to the racial comments hurled against her b y the Sandra and leaves feeling insulted. The two car thieves Anthony and peter proceed to the SUV, discussing issues of racism and country music. While in the discussion, they hit an Asian man, the two are puzzled by the event and instead of helping the injured victim they dump him in front of a hospital. Under normal circumstances, one would expect immediate first aid to the car accident victim or being rushed to hospital for medical check-up. The negligent attitude by peter and Anthony can be linked to the racial difference between them and the Asian man. This shows how bad racial prejudice can be; to an extent that on looses the human feelings towards fellow humans just because they belong to a different race. When eventually the two return the car to the owner, Mr. Lucien, he refuses to take the car because it had bloodstains. This was a clear indication that gang targeted members of different races but not their fellow blacks. Their target is clear prejudice directed towards other races. There is general hatred for the racist cop by Shaniqua. Ryan pays Shaniqua a visit to explain the condition of his ailing father and how he badly requires a transfer. Because of the hatred that Shaniqua has towards Ryan, she refuses to grant her the transfer arguing that the health cover on offer does not take care of the fathersââ¬â¢ case. Ryan is annoyed by her objection and tries to tell her how the now ailing father fought tirelessly for affirmative action to grant equal rights to the blacks as the minority group. Shaniqua reveals that he could only accept to the request if it was the father who had come but not Ryan. This episode is an illustration of the preju dice that those holding office position have to people who do not belong to their race. Services even in public offices are offered with divided loyalty based on which race one belongs. This are samples of new forms of racism that are currently replacing the older forms of racial prejudice (Ebert 2009, p. 232). The racist cop Ryan seems to have been noted by many people who are equally waiting for a chance to revenge against his behavior. When Cameron goes to the studio where he works, the producer who is a white claims one of the black actors in the movie is not acting as a black. The producer insists the black actor should act properly like a black person. Because of this, the producer suspends the production of the movie and orders Cameron to re-shoot the movie again. The treatment he receives from the white producer leaves him disgusted and annoyed by the comments made by the producer about the blacks. Two racial issues are clear from this episode, one is the perception that t he white producer has about how the blacks should act in the movie and the attitude that he has towards Cameron as to force him to reshoot the movie to look like black. At the same incidence, Cameronââ¬â¢s wife Christine arrives and reminds him of the previous instance in the hands of the racist cop. Memories of the act makes Cameron more annoyed and he leaves her crying. Farhad refused to pay Daniel a black man for fixing the lock to his door after he told him he needed a new door. Daniel left disgusted and a few days later, Farhadââ¬â¢s shop was wrecked and labeled with racial impressions in graffiti. Farhad was so much annoyed by the act and set out to find the locksmith. He went to an insurance company for compensation but was turned away the company terming it an act of negligence. Farhad proceeded to the locksmith company demanding to be told the name of the fellow who had fixed the lock to his shop but the company refused to disclose the name to him. Farhad wanted to launch revenge to Daniel for the racist illustrations and the wrecking of his shop. Racial prejudice is revealed in the decision that the locksmith takes to put racist illustrations on the shop as revenge against the owner refusing to pay him. There are definitely many legal steps Daniel would have taken to demand payment from Farhad, instead he choose to hurt Farhad by the graffiti illustrations on his shop. Discussion When Christine was involved in an accident, Ryan was on patrol and ran to the scene of accident to assist the casualties. Christine was trapped in the crashed car and crying out for help, when Christine saw Ryan as one among those who had come to her rescue, she screamed at him. She remembered the ordeal she underwent in the hands of Ryan and felt he could easily do the same again. Ryan was more human this time and assisted together with others at the scene to pull her out of the nearly burning wreck. This time Christine was more grateful to Ryan for the effort to a ssist her out of the nearly burning wreck. As Hilliard (2009, p. 147) points out, racism is more of a personal responsibility and should not be generalized to a group. The impact of racism is felt more by those who have ever been victims of the act from others. Another scene that depicts the role of African Americans in racial prejudice is brought out when Cameron was driving as stopped at a road sign. Peter and Anthony the two black carjackers pulled over to carjack him, not realizing that he was one of them. A fight ensues between them which attract the nearby cops to the scene. Cameron and Anthony take off together in the car which the cops decide to give a chase. Their car is eventually cornered and Cameron comes out threatening the cops, Anthony instead hides in the back seat. The two are arrested by the police for investigation but later released from the copsââ¬â¢ custody. Cameron is annoyed by Anthony terming him an embarrassment. It is clear in this episode the prejudic e Anthony and peter have against the whites, their attack on Cameron a fellow black was a mistaken. They targeted to carjack at least a person of different race but not one of their own. When they are cornered Cameron comes out to threaten the white cops but Anthony instead hides in the car. Cameron is annoyed at how Anthony can not come out to assist him hit back at the cops. Peter and the cop Hansen meet in a controversial scene; peter picked up Hansen and was seen to be in a friendly mood chatting heartily with him. A problem later ensues when peter points at the statute on Hansenââ¬â¢s dashboard. It was a statute of sir Christopher, instead peter pulls out his statute from the pocket and tries to justify how his was more superior than that of Hansen. Hansen is annoyed by peterââ¬â¢s reaction to his statute and shoots him dead. The movie begins with characters that seem to be all independent but only reveal their dependence towards the end of the movie. After shooting pe ter, Hansen abandoned his car and set it ablaze, this may be associated likely to the guilt that he had after shooting and damping peterââ¬â¢s body. Cameron comes across the burning car and instead of trying some effort that could stop the fire; he threw a piece of wood into the fire. This was intended to even fan the fire more. The act was because the car belonged to Hansen who was a white cop. On visiting the chop shop, Anthony met a group of illegal immigrants to whom Lucien offered $500 each, Anthony refused the money and instead offered to drop off the immigrants to Chinatown, he also gave one of them some money to buy himself food. This act towards the immigrant left him more redeemed. Conclusion Haggis succeeded in painting the picture of human weakness in the face of racial or ethnic differences. The modern America comprises of different racial groups from all over the world who have migrated and settled in search of opportunities. Others were taken there during the days of slavery. There are also illegal immigrants who continue to flock in from various parts of the world seeking for better opportunities or running away from problems in their countries. The differences based on race, culture and ethnicity often brew serious tensions among the various groups. It is worthy noting that racism is not a problem of one particular group or race but a problem of humanity. There is no one group that is a victim forever just as there is no group that is a perpetual creator of racism. Human beings tend to cluster one another according to the similarities they have in terms of race and ethnicity. Some of the scenes in the Movie Crash can be offending to viewers from the different various racial groups but holds a lot of truth (Garret 2007, p. 126).
